Entheias has developed a proprietary therapeutic vaccine platform aimed at achieving functional cure in chronic hepatitis B (HBV), a disease affecting approximately 254 million people globally and responsible for nearly one million deaths annually from cirrhosis and liver cancer. Current standard therapies, including nucleos(t)ide analogues and antiviral suppression strategies, rarely achieve a durable functional cure, while emerging therapeutic vaccines continue to leave a substantial percentage of patients unresponsive to treatment.
The Entheias platform is specifically designed to address this unmet need by targeting the root immunological causes behind therapeutic vaccine non-response. Existing HBV immunotherapies and combination regimens have demonstrated promising efficacy in responder populations, yet approximately 40% of patients fail to mount sufficient immune activation to achieve meaningful clinical outcomes. Entheias seeks to convert these non-responders into responders through a novel mechanism of action built on validated immunological principles with supporting in vivo precedent from cancer immunotherapy research.
The platform is engineered to work synergistically with current and emerging HBV treatment backbones, including antisense oligonucleotides (ASOs), siRNA therapies, checkpoint inhibitors, and PEG-interferon approaches, rather than competing directly with them. By complementing established antiviral suppression methods with targeted immune restoration, the technology aims to significantly expand overall functional cure rates across the broader patient population.
In addition to its therapeutic focus, the platform incorporates rapid optimization and iteration capabilities, enabling accelerated candidate refinement and lower-cost preclinical validation. The underlying technology is also designed as an extensible immunotherapy platform with potential applications beyond HBV into other chronic viral infections.
